Evaluating the prognostic impact of subclonal FLT3-ITD mutations in acute myeloid leukemia
One of the activities that we will give our highest priority to in the next weeks is a project evaluating the prognostic impact of subclonal FLT3-ITD mutations in acute myeloid leukemia; a retrospective multicenter study on behalf of the EHA AML-SWG. In 2023, this project was selected for funding with an EHA “initiating grant”. 6 centres across Europe and UK are participating in this project.
A total number of n=908 FLT3-ITD mutated patients have been identified from a cohort of more than 10.000 AML diagnosed between 2017-2022, intensively treated with standard chemotherapy or less intensive treatment. The data has been analyzed, and the first results were presented at the last ASH meeting.
A manuscript is currently in preparation and is planned to be submitted in April.
Validation of an appropriate blast threshold for AML definition
The validation of an appropriate blast threshold for AML definition: MDS/AML (10-19%) vs AML (≥20%) category, as recently proposed by the International Consensus Classification (ICC). The project is led by Jordi Esteve.
The project was recently approved by HARMONY. Using the HARMONY platform, more than 1000 patients have been identified for the different blast threshold groups.
Additional datasets are currently being compiled for the validation cohort, and initial analyses are underway.
Observational study on AML in pregnancy
Observational study on AML in pregnancy: treatment and maternal and fetal outcomes: a retrospective and prospective study under the umbrella of the EHA SWG AML. This project is led by Nicola Fracchiolla and Francesco Passamonti from the Hematology Unit, Ospedale Maggiore Policlinico in Milano, Italy.
The study protocol and the data dictionary have been finalized and circulated within the SWG AML as well as in centers in Italy and beyond.
The SWG AML is currently exploring regulatory issues such as informed consent, data transfer agreement as well as approval by the respective Ethic Committee.
SWG collaboration
Joint project between SWG Infections in Hematology and the SWG aiming to gather data on infections in AML patients undergoing induction, consolidation, and salvage chemotherapy across European Hematology Association (EHA) institutions.
The primary objective is to assess the incidence and type/spectrum of bacterial, fungal, and viral infections in AML patients at the defined time points. An anonymized electronic database will be used to collect data on demographics, epidemiology, admission details, disease status, treatments, and infection events related to AML.
The protocol of this study has been finalized and approval by the Ethic Committee of Catholic University in Roma has already been obtained. The SWG AML is currently exploring regulatory issues such as informed consent, data transfer agreement as well as approval by their respective Ethic Committees.
Additional aims:
- To further develop the ongoing AML projects, as well as defining new projects.
- Harmonization of molecular techniques between the research groups e.g. in close collaboration with ELN AML Work package 5 by developing/up-dating MRD guidelines/recommendations and performing laboratory cross-validations.
- The consideration of the implications of high throughput sequencing technologies and measurable residual disease assessment in clinical trial design.

The 2nd EHA-SWG Scientific Meeting on MDS/MPN/AML: Commonalities and Differences of Myeloid Neoplasms

- Collaborating SWGs: EHA Specialized Working Group on Myelodysplastic Syndromes (MDS), EHA Specialized Working Group on Myeloproliferative Neoplasms (MPN), and EHA Specialized Working.
- The 2nd EHA-SWG Scientific Meeting on MDS/MPN/AML will focus on the biological and clinical commonalities and differences across myeloid neoplasms.
- The program will address disease pathogenesis, clonal evolution, and molecular features, as well as emerging therapeutic strategies and precision medicine approaches.
- It will also highlight overlap syndromes and promote interdisciplinary discussion and collaboration among experts in the field.
